Herb Trimpe and Jack Abel's The Incredible Hulk #180 page was sold for $657,250 at Heritage Auctions.
Todd McFarlane's cover art for The Amazing Spider-Man #328 sold for the same price in 2012.
The sale marks the record for a single piece of interior comic art.
"We knew when this artwork surfaced that was, without doubt, one of the most significant pieces of original comic art ever drawn," said vice president of Heritage Auctions Todd Hignite.
"It has now brought a final price realised commensurate with that status."
Trimpe gave the artwork from the 1974 Marvel Comics issue to the seller in 1983 when he was a teenager. A portion of the sale will be donated to the Hero Initiative.
