Hamill, who originally played the villain in the '90s CBS series, is back to help Barry Allen/The Flash and Joe West track down various copycat Tricksters creating havoc in the city.
The trailer of upcoming villains also features Spartacus' Liam McIntyre as Weather Wizard, as well as the return of Captain Cold (Wentworth Miller) and Heat Wave (Dominic Purcell), accompanied by Snart's sister Lisa (Peyton List).
The Stars Wars star will appear in the show's March 31 episode.
Executive producer Andrew Kreisberg recently described the actor's return as if "Christmas and Chanukah came early".
"We are beyond humbled and excited to have him joining us for another round of deadly trickery!" he said.
The CW are reportedly working on a new Arrow and Flash spinoff, starring Brandon Routh (Ray Palmer / The Atom), Caity Lotz (Sara Lance / Canary), Miller (Leonard Snart / Captain Cold) and Victor Garber (Dr Martin Stein / Firestorm).
The Flash airs on The CW in the US. Sky1 airs the show in the UK, Tuesday nights at 8pm.
